Rho family GTPase-dependent immunity in plants and animals

چکیده
In plants, sophisticated forms of immune systems have developed to cope with a variety of pathogens. Accumulating evidence indicates that Rac (also known as Rop), a member of the Rho family of small GTPases, is a key regulator of immunity in plants and animals. Like other small GTPases, Rac/Rop GTPases function as a molecular switch downstream of immune receptors by cycling between GDP-bound inactive and GTP-bound active forms in cells. Rac/Rop GTPases trigger various immune responses, thereby resulting in enhanced disease resistance to pathogens. In this review, we highlight recent studies that have contributed to our current understanding of the Rac/Rop family GTPases and the upstream and downstream proteins involved in plant immunity. We also compare the features of effector-triggered immunity between plants and animals, and discuss the in vivo monitoring of Rac/Rop activation.

Vibrio parahaemolyticus inhibition of Rho family GTPase activation requires a functional chromosome I type III secretion system.
Vibrio parahaemolyticus is a leading cause of seafood-borne gastroenteritis; however, its virulence mechanisms are not well understood. The identification of type III secreted proteins has provided candidate virulence factors whose functions are still being elucidated. Genotypic strain variability contributes a level of complexity to understanding the role of different virulence factors. The ab...
متن کاملIntercalating Arabidopsis Leaf Cells: A Jigsaw Puzzle of Lobes, Necks, ROPs, and RICs
Intercalation of cells is an evolutionarily conserved strategy used for a variety of developmental processes in animals. In this issue of Cell, Fu et al. have uncovered an elaborate Rho GTPase-mediated mechanism by which cytoskeletal-dependent intercalation of Arabidopsis leaf cells is achieved, suggesting that conserved Rho GTPase signaling pathways may similarly regulate tissue morphogenesis ...
متن کاملArabidopsis RopGAPs are a novel family of rho GTPase-activating proteins that require the Cdc42/Rac-interactive binding motif for rop-specific GTPase stimulation.
The plant-specific Rop subfamily of Rho GTPases, most closely related to the mammalian Cdc42 and Rac GTPases, plays an important role in the regulation of calcium-dependent pollen tube growth, H(2)O(2)-mediated cell death, and many other processes in plants. In a search for Rop interactors using the two-hybrid method, we identified a family of Rho GTPase-activating proteins (GAP) from Arabidops...
متن کاملInactivation of rho GTPases by statins attenuates anthrax lethal toxin activity.
Anthrax lethal factor (LF), secreted by Bacillus anthracis, interacts with protective antigen to form a bipartite toxin (lethal toxin [LT]) that exerts pleiotropic biological effects resulting in subversion of the innate immune response. Although the mitogen-activated protein kinase kinases (MKKs) are the major intracellular protein targets of LF, the pathology induced by LT is not well underst...
متن کاملLocalization of a Rho GTPase Implies a Role in Tip Growth and Movement of the Generative Cell in Pollen Tubes.
The Rho family GTPases function as key molecular switches, controlling a variety of actin-dependent cellular processes, such as the establishment of cell polarity, cell morphogenesis, and movement in diverse eukaryotic organisms. A novel subfamily of Rho GTPases, Rop, has been identified in plants. Protein gel blot and RNA gel blot hybridization analyses indicated that one of these plant Rho GT...
